GUI actually stands for Graphical User Interface...in regular ole terms it means that the screen shows a "picture" of a control panel, or instrument that you are working with instead of a "text' only screen. With a GUI interface if you wanted to change the setting of say, a filter on an oscillator bank you would click on the filter knob or slider that is shown on the picture of the control panel, as opposed to going thru an edit process and finding in a text style file the filter paramaters. Just makes working with VSTs (or any computer program for that matter) way easier.
As to just using a monitor/keyboard/mouse instead of a laptop to config the Muse...the advantage of a laptop is some programs, like the Doepfer's editor must be run on a computer/laptop, as it has no smarts to it and looks for the PC to prvide that. Remember that the muse can go without a computer and just use a monitor because it IS a computer.
