Unfortunately, no. The Exp/Overdrive knob sends out the MIDI CC# that you have defined to be expression, and it is always either in parallel with the (foot) expression pedal input, or disabled. It cannot be re-programmed to do anything else. You could use the function for something else (Amp Gain?), but you have to disable MIDI expression response to get there, which seems like a bad idea to me.

There are no "spare" functions on the CX-3, but there is another way to get at the wheel brake: Press Global, "8", and then "-". You should see "Wheel Brake" on the display. Press Write/Enter to engage the brake, and press it a second time to cancel the effect.
